Can you first give us the hints the posting guide asked you for?  What OS, 
what sort of GUI, what sort of application?

If you look on CRAN/BioC you will see that most people use Tcl/Tk, a few
use RGtk2, and you can also embed R into an alternative front-end (we've 
done that via Visual Basic, some people link to Excel ....).
